The Pilgrims' 12th consecutive campaign at Step 2 level will get under way (kick-off 3.00pm) as Craig Elliott's side look to go one better than the last completed season when they suffered promotion final anguish at the hands of Altrincham.
This time around, Elliott will be hoping to field his full quota of summer signings – although he will check on the fitness of midfield trio Shane Byrne, Andi Thanoj and Paul Green, who all missed out against Grimsby Town last weekend.
Scott Duxbury (ankle) and Jay Rollins (knee) will definitely be missing after sustaining injuries during the Mariners clash.
Opponents Spennymoor, now managed by Tommy Miller, have ex-Pilgrims Brad Abbott, Jason Kennedy and Frank Mulhern within their ranks.
The sides have not met since November 2019, when Dominic Knowles was on target for the Pilgrims in a 2-1 defeat at the Brewery Field.
