The Pilgrims entertain the Magpies' youngsters (kick-off 7.30pm) in the third group stage outing at the Jakemans Community Stadium.
A 4-3 opening night defeat to Leeds United was followed by a 1-0 victory over Sunderland – with a further fixture against Middlesbrough still to unfold in the New Year.
The Magpies have beaten FC Halifax Town and drawn with Tamworth in their opening fixtures.
United manager Graham Coughlan will be without the services of cup-tied duo Connor Teale and Lennon Wheeldon, who have already played in the competition this season.
Tony Weston is expected to make his first appearance since mid-October, following his recovery from a broken metatarsal.
