The Victoria Park encounter (kick-off 3.00pm) is United's first since September 2006 when Coca-Cola League Two points were at stake.
Pools were 2-1 victors at the Jakemans Community Stadium earlier in the season, despite the Pilgrims holding a half-time lead through Jacob Hazel.
After a season of managerial upheaval, Hartlepool are now managed by Anthony Limbrick, with Darren Sarll and Lennie Lawrence also having spent time in the hotseat.
They sit nine points above the Pilgrims ahead of kick-off, although Graham Coughlan's side have two matches in hand.
United will check on the fitness of midfield duo Frankie Maguire and Martin Woods, who both sat out Tuesday's 3-2 win at Rochdale through injury.
