The Pilgrims, with three draws and three defeats to their name inside their opening six league fixtures, face Alun Armstrong's Quakers side.
Darlington have recorded home wins over Gloucester City and Farsley Celtic to date – as well as securing points on the road at Chorley and Spennymoor Town.
United will be without the services of suspended striker Jaanai Gordon – and will check on the fitness of captain Luke Shiels, who limped out of Bank Holiday Monday's 1-1 draw against Kettering Town.
Manager Paul Cox will also assess whether Zak Mills, Brad Abbott and Jake Wright are fit enough to return – with Tom Platt and Jordan Burrow both still nursing ankle problems.
Last season's corresponding fixture ended 2-1 to the hosts, despite Danny Elliott's second half penalty.
