Culverhouse and his trusted assistants Paul Bastock and Joe Simpson will take charge of the Pilgrims for the first time against Billy Heath's Reds.
United will be without suspended duo Lebrun Mbeka and Jaanai Gordon, with Culverhouse set to check on the fitness of captain Luke Shiels and Salford City loanee Colin Oppong.
While United are still searching for their first win of the season at the eighth attempt, Alfreton have won two and drawn four of their opening seven matches.
Heath's squad includes a number of ex-Pilgrims, including George Willis, Fraser Preston and Dayle Southwell.
Last season's corresponding fixture ended 3-2 in Alfreton's favour, in what turned out to be Craig Elliott's final match in charge.
